CARA KERJA OBAT :
Amoksisillin bersifat bakterisid, efektif terhadap bakteri gram negatif seperti Haemophilus influenza, Escherichia coli, Proteus mirabilis Neisseria gonorrhoeae dan bakteri gram positif seperti Streptococci (termasuk Streptococci faecalis), B. pneumoniae non penicillinase-producing Staphylococci. Prinsip kerjanya adalah menghambat sintesa dinding sel bakteri.

INDIKASI :
Untuk pengobatan infeksi karena mikroorganisme gram positif dan gram negatif yang peka terhadap Amoksisillin, misalnya : infeksi pernafasan bagian bawah, saluran kemih, saluran pencernaan, juga infeksi-infeksi kulit dan jaringan lunak.

KONTRA INDIKASI :
Penderita yang sensitif terhadap Penisilin.

EFEK SAMPING :
- Gangguan pada saluran pencernaan misalnya : Nausea, vomitus, diare, glositis dan stomatitis.
- Alergi misalnya : Skin rashes, urtikaria, reaksi makulo papular, eritema, pruritis, angioedema, serum sickness, kadang-kadang dapat terjadi reaksi anafilaksis.

PERINGATAN DAN PERHATIAN :
- Keamanan pada wanita hamil belum diketahui dengan pasti.
- Kemungkinan timbul super infeksi yang disebabkan oleh Candida, Pseudomonas, Enterobacter pada pemakaian jangka lama atau dosis tinggi.

INTERAKSI OBAT :
Pemberian bersama Probenecid dapat mempertinggi kadar Amoksisillin dalam serum darah, karena Probenecid memperlambat sekresi Amoksisillin oleh ginjal.

ATURAN PAKAI :
Dewasa & anak-anak > 20 kg : 750 - 1500 mg/hari, tiap 8 jam
Anak-anak < 20 kg : 20 - 40 mg/kg BB/hari, tiap 8 jam
Atau menurut petunjuk dokter.

COMPOSITION:
- SUPRAMOX 500 tablet, each tablet contains:
Amoxicillin trihidrate equal to Amoxicillin 500 mg
- SUPRAMOX 500 capsule, each capsule contains:
Amoxicillin trihidrate equal to Amoxicillin 500 mg
- SUPRAMOX Dry Suspension, each 5 mL contains:
Amoxicillin trihidrate equal to Amoxicillin 125 mg
- SUPRAMOX Forte Dry Suspension, each 5 mL contains:
Amoxicillin trihidrate equal to Amoxicillin 250 mg
- SUPRAMOX Drops, each 1 mL contains:
Amoxicillin trihidrate equal to Amoxicillin 100 mg

MECHANISM OF ACTION:
Amoxycillin is a highly potent, broad-spectrum penicillin. It acts by inhibiting cell wall synthesis. It is active against both gram-positive and gram-negative microorganisms, eg: Escherichia coli, Haemophilus influenzae, Proteus mirabilis, Neisseria gonorrhoeae, Streptococcus (including Streptococcus faecalis), Streptococcus pneumoniae, non-penicillinase-producing Staphylococcus.

INDICATIONS:
Supramox is usefull in the treatment of infections caused by amoxicilin-susceptible organisms:
- Respiratory Tract Infections: Upper airways and ENT infections, lower airways infections (eg. Acute and chronic bronchitis, pneumonia, lung abscesses, pertusis.
- Urogenital Infections: Acute and chronic pyelonephritis, pyelitis, prostatitis, epididymitis, cystitis, urethritis, asymptomatic bacteriuria during pregnancy, gonorrhoea.
- Gynaecologic infections (septic abortion, adnexitis, endometritis, etc).
- Gastrointestinal Infections: Typhoid fever, paratyphoid, particularly if complicated by septicaemia, control of Salmonella carriers, shigellosis, infection of the biliary system (cholangitis, cholecystitis).
- Skin and soft tissue infections.
- Leptospirosis.

CONTRA-INDICATIONS:
Hypersensitivity to penicillins.

ADVERSE REACTIONS:
- Occasional transient side effects are mostly gastrointestinal (nausea, vomiting, diarrhoea, glossitis, stomatitis).
- Rarely, hypersensitivity reactions like urticaria, maculo-papular reaction, erythema, pruritus, angioedema, serum sickness, severe systemic reaction (anaphylactic shock).

PRECAUTIONS AND WARNINGS:
- There is no current evidence to suggest any embryotoxic, teratogenic or mutagenic effects of amoxycillin during pregnancy.
- During long-term treatment attention should be paid to the potential overgrowth of resistant microorganisms and fungi.

DRUG INTERACTIONS:
The concomitant administration of probenecid produces sustained and higher plasma levels by suppressing renal elimination.

DOSAGE AND ADMINISTRATION:
Adult and children >20 kg: 750-1500 mg daily, every 8 hours.
Children <20 kg: : 25-50 mg/kg body weight daily, every 8 hours.
